Cotyledonary node, axillary node, hypocotyls, epicotyls of young seedlings of Sesbania drummondii were cultured on Murashige and Skoog's (MS) and Gamborg's (B5) basal media supplemented with different concentrations and combinations of NAA (0.1-2.0 mg/L); 6-benzyl amino purine (0.5-2.5 mg/L) and kinetin (3.0-5.0 mg/L). After 4-6 weeks, profuse growth of callus was observed on MS medium with the combination of 1.5 mg/L NAA and 2.0 mg/L BAP. Development of callus was vigorous on MS medium with 1-2 mg/L 2, 4-D and 5 mg/L kinetin. Subculture of callus on MS medium (without plant growth regulators) induced embryoid-like globular structures on callus surfaces. Sub-culturing on different concentrations and combinations of 2, 4-D (0.5-1.5 mg/L) and kinetin (3-5 mg/L) produced several root-like hairy structures. Callus established on LS basal media supplemented with .1-.3 mg/L 2, 4-D also had embryoid-like globular or tubular structures. Attempts are being made to achieve shoot regeneration from callus and globular embryoids.
